Sample size for the assessment of soybean inbred populations Colombari Filho, José Manoel Geraldi, Isaias Olívio

No melhoramento genético de plantas é muito importante o conhecimento do tamanho da amostra para avaliar as populações. Uma amostra pequena reduz a chance de seleção de genótipos superiores, enquanto que uma amostra grande pode acarretar aumentos desnecessários de custo e trabalho. Uma população composta de 192 linhagens de soja foi dividida aleatoriamente em grupos de 24 linhagens, que foram avaliadas para a produção de grãos em oito experimentos em blocos ao acaso. Foram realizadas análises de variância para os oito experimentos e para o agrupamento destes, obtendo-se análises de variância com 24, 48, 72, 96, 120, 144, 168, and 192 linhagens. Observou-se que conforme o tamanho da amostra aumentou, os intervalos de confiança das estimativas de parâmetros diminuíram, estabilizando com amostras de 144 linhagens. Portanto, uma amostra apropriada para a avaliação de uma população endogâmica de soja deve conter aproximadamente 150 linhagens.

In plant breeding programs, the knowledge about the appropriate sample size for the evaluation of populations is very important. A small sample reduces the chance of selecting superior genotypes, whereas a very large sample may lead to unnecessary increases in cost and labor. A population consisting of 192 soybean lines was divided in groups of 24 lines, which were assessed for grain yield in eight randomized complete block experiments. Analyses of variance were performed for each experiment as well as for groups of experiments, resulting in analyses of variance consisting of 24, 48, 72, 96, 120, 144, 168, and 192 lines. As the sample size increased, the width of confidence intervals of parameter estimates decreased, stabilizing with samples of 144 lines. Therefore, an appropriate sample size for the evaluation of soybean inbred populations should contain about 150 lines.

A glutathione s-transferase confers herbicide tolerance in rice Hu, Tingzhang

Plantas glutationa s-transferase (GSTs) têm sido foco da atenção devido ao seu papel na detoxificação de herbicida. OsGSTL2 é uma glutationa s-transferase, gene da classe lambda de rice (Oryza sativa L.). Plantas transgênicas de arroz superexpressando OsGSTL2 foram geradas de "calus" de arroz via sistema de transformação de Agrobacterium, e foram avaliadas por uma combinação de resistência a higromicina, PCR e análise de "Southern blot". Nos tecidos vegetativos das plantas transgênicas, a superexpressão de OsGSTL2 não somente incrementou os níveis de transcritos OsGSTL2, mas também a expressão de GST e GPX, enquanto reduziu superóxido. Plantas transgênicas também mostraram mais alta tolerância a glifosato e clorosulfuron, os quais frequentemente contaminam solos agrícolas. Essas descobertas demonstram o papel de detoxificação de OsGSTL2 no crescimento e desenvolvimento de plantas de arroz. A partir delas será possível também aplicar tais resultados a outros cultivos para desenvolvimento de tolerância a herbicida e para limitar a contaminação com esse agrotóxico na cadeia alimentar.

Abstract in English:

Plant glutathione S-transferases (GSTs) have been a focus of attention due to their role in herbicide detoxification. OsGSTL2 is a glutathione S-transferase, lambda class gene from rice (Oryza sativa L.). Transgenic rice plants over-expressing OsGSTL2 were generated from rice calli by the use of an Agrobacterium transformation system, and were screened by a combination of hygromycin resistance, PCR and Southern blot analysis. In the vegetative tissues of transgenic rice plants, the over-expression of OsGSTL2 not only increased levels of OsGSTL2 transcripts, but also GST and GPX expression, while reduced superoxide. Transgenic rice plants also showed higher tolerance to glyphosate and chlorsulfuron, which often contaminate agricultural fields. The findings demonstrate the detoxification role of OsGSTL2 in the growth and development of rice plants. It should be possible to apply the present results to crops for developing herbicide tolerance and for limiting herbicide contamination in the food chain.

A obtenção de correlações e os efeitos diretos e indiretos dos componentes do rendimento com a produtividade de grãos são importantes para a seleção de parentais e populações segregantes promissoras. O objetivo da pesquisa foi estimar as correlações fenotípicas e genotípicas e o desdobramento dos efeitos diretos e indiretos dos componentes de rendimento com o rendimento de grãos de20 genótipos de feijão-caupi (Vigna unguiculata). O delineamento experimental foi em blocos casualizados, com 20 tratamentos e 4 repetições. Os caracteres demonstraram baixa à moderada possibilidade de ganho por seleção indireta, havendo maior probabilidade de sucesso ao aliar múltiplos caracteres e genótipo de melhor performance.

Obtaining correlations and direct and indirect effects of yield components is important for the selection of promising parental and segregating populations. The objective of this research was to estimate the genotypic and phenotypic correlations, and to analyze the direct and indirect effects of yield components on grain yield in 20 cowpea (Vigna unguiculata) genotypes. The experimental design was in randomized blocks with 20 treatments and 4 replications. The character showed low to moderate possibility of gain from indirect selection, with greater possibility for success when joining multiple character and a genotype of better performance.

O objetivo desse estudo foi selecionar progênies de café produtivas e com resistência à ferrugem em dois locais do Estado do Paraná (Brasil). Os experimentos de campo foram instalados no delineamento experimental em blocos ao acaso nos município de Itaguajé e Congonhinhas. Foram avaliadas as características produção, vigor vegetativo e resistência à ferrugem em nove progênies de café arábica e em três cultivares padrões. Vários genótipos derivados do "Sarchimor" e do "Catucaí" apresentaram plantas suscetíveis. Três cafeeiros do germoplasma Sarchimor e uma progênie "F6 de Catuaí x (Catuaí x cafeeiro da série BA-10)" foram selecionados para avanço de geração e têm potencial para se tornarem novas cultivares, pois apresentaram produção maior que 'IAPAR 59' e 'Tupi IAC 1669-33' e muitas plantas apresentaram resistência completa à ferrugem.

The aim of this study was to select high-yielding coffee progenies with resistance to coffee leaf rust for the State of Paraná (Brazil). Field experiments in a randomized block design were carried out in Itaguajé and Congonhinhas. Yield, vegetative vigor and rust resistance of nine progenies of Arabic coffee and three check cultivars were evaluated. Many genotypes derived from "Sarchimor" and "Catucaí" were susceptible. Three coffee genotypes of Sarchimor germplasm and the F6 generation of genotype "Catuaí x (Catuaí x BA-10 coffee)" were selected to advance generations and are promising to become new cultivars for being higher-yielding than 'IAPAR 59' and 'Tupi IAC 1669-33' and having a large number of plants with complete rust resistance.

Selection of coffee progenies for resistance to nematode Meloidogyne paranaensis in infested area Salgado, Sonia Maria Lima Rezende, Juliana Costa de Nunes, José Airton Rodrigues

Objetivou-se neste trabalho selecionar progênies de Coffea arabica para resistência à M. paranaensis em área cafeeira infestada usando a abordagem de modelos mistos de Henderson. Quarenta e um genótipos foram selecionados no Banco Ativo de Germoplasma de Café de Minas Gerais e avaliados individualmente quanto ao diâmetro de caule, número de ramos plagiotrópicos, caracterização da reação dos genótipos ao nematoide e produção por planta. Houve variabilidade genética entre os genótipos estudados para todas as características avaliadas, e entre as populações estudadas para as características produção e caracterização da reação dos genótipos ao nematoide, indicando possibilidades de obtenção de ganhos genéticos pela seleção nesta população. As correlações genotípicas entre todas as características estudadas foram significativas. Cafeeiros da população de Híbrido de Timor UFV 408-01 e progênies F3 derivadas do cruzamento entre Catuaí Vermelho e Amphillo MR 2161 foram os mais promissores em área infestada por M. paranaensis.

The purpose of this study was to select Coffea arabica progenies for resistance to M. paranaensis in an infested coffee growing area using Henderson's mixed model methodology. Forty-one genotypes were selected at the Coffee Active Germplasm Bank of Minas Gerais, and evaluated in regard to stem diameter, number of plagiotropic branches, reaction to the nematode, and yield per plant. There was genetic variability among the genotypes studied for all the traits evaluated, and among the populations studied for yield and reaction to the nematode, indicating possibilities for obtaining genetic gains through selection in this population. There was high rate of genotypic association between all the traits studied. Coffee plants of Timor Hybrid UFV408-01 population, and F3 progenies derived from crossing Catuaí Vermelho and Amphillo MR 2161 were the most promising in the area infested by M. paranaensis.

Cana-de-açúcar possui flores hermafroditas, no entanto, autofecundação e polinização cruzada podem ocorrer, resultando em progênies autofecundadas ou híbridas. O objetivo do presente estudo foi analisar a paternidade de progênies provenientes de cruzamentos biparental, para identificar híbridos verdadeiros ou progênies formadas a partir de pólen de origem desconhecida. Setenta e seis progênies de quatro cruzamentos foram analisadas usando três marcadores moleculares microssatélites (SSR) altamente polimórficos. As progênies mostraram moderada similaridade genética e foram agrupadas em quatro distintos grupos, conforme os cruzamentos. A transmissão dos alelos dos parentais para a descendência foi claramente observada, em que não foram observados indivíduos autofecundados, somente híbridos verdadeiros ou progênies resultante da fecundação de pólen incomum a ambos os parentais. Os resultados mostraram que existe contaminação com pólen de genitores desconhecidos em cruzamentos em cana-de-açúcar, sugerindo que erros no pedigree podem ocorrer.

Abstract in English:

Sugarcane has hermaphrodite flowers, however, selfing and cross pollination may occur, resulting in selfed or hybrid progeny. The aim of this study was to analyze the paternity of progenies from biparental crosses, in order to identify true hybrids or progenies originating from pollen of unknown origin. Seventy-six progenies from four crosses were analyzed using three highly polymorphic microsatellite markers (SSR). Progenies showed moderate genetic similarity and were grouped into four distinct groups, according to the crosses. Transmission of alleles from parents to offspring was clearly observed, in which selfed individuals were not observed, and only true hybrids or progeny resulting from fertilization with pollen uncommon to both parents were. Results showed that there was contamination with pollen from unknown parents in sugarcane crosses, suggesting that errors in the pedigree may occur, and adjustment in the crossing procedure would decrease progenies from pollen of unknown origin.

Micropropagation and callogenesis in Mandevilla guanabarica (Apocynaceae), an endemic plant from Brazil Cordeiro, Sandra Zorat Simas, Naomi Kato Henriques, Anaize Borges Sato, Alice

Abstract in Portuguese:

Mandevilla guanabarica é uma planta endêmica do Brasil, com potencial farmacológico e ornamental inexplorados. Este estudo estabeleceu um eficiente meio de cultura para manutenção da planta in vitro e eficazes protocolos de regeneração e calogênese a partir de diferentes explantes. O meio para plantas lenhosas suplementado com o dobro da concentração de boro (WPM B) e com 2,27 µM de thidiazuron ou 0,49 µM 2-isopenteniladenina proporcionou taxas de multiplicação maiores que 1:6. Os brotos foram 100% enraizados em WPM B, com 83% de sobrevivência após aclimatização. Na calogênese, o meio MS suplementado com altas concentrações de picloram ou ácido 2,4-diclorofenoxiacético produziu, respectivamente, calos não morfogênicos friáveis ou compactos. Este protocolo de micropropagação permite a produção de mudas de M. guanabarica para uso ornamental, comercial e com finalidades de conservação; os calos podem ser utilizados para o estabelecimento de culturas em suspensão na prospecção de compostos bioativos.

Abstract in English:

Mandevilla guanabarica is an endemic plant from Brazil, with pharmacological and ornamental potential, both unexplored. This study established the best culture medium for in vitro plant maintenance, efficient protocol for its regeneration, and callogenesis from different explants excised from in vitro-grown plants. Woody plant medium with double boron concentration (WPM B) plus 2.27 µM thidiazuron or 0.49 µM 2-isopentenyladenine provided multiplication rates higher than 1:6. Shoots were 100% rooted on WPM B. After acclimatization, plants showed 83% survival. For callogenesis, the use of MS media supplemented with high concentrations of picloram or 2,4-dichlorophenoxyacetic acid produced, respectively, friable or compact non-morphogenic calluses from different types of explants. This micropropagation protocol allows the production of seedlings of M. guanabarica for ornamental or commercial uses, and for conservation purposes; calluses can be used to establish suspension cultures in prospecting for bioactive compounds.
Genetic analysis of the naked trait in panicles of hexaploid oat Valentini, Ana Paula Fontana Pellizzaro, Kelly Pacheco, Marcelo Teixeira Federizzi, Luiz Carlos

Abstract in Portuguese:

O objetivo deste trabalho foi estimar o número de genes que controlam o caráter nuda e o modo de expressão desta característica em panículas de aveia branca hexaplóide. Genitores e população segregante (nas gerações F2 e F3) foram avaliadas quanto à presença e distribuição de grãos nudas em panículas de plantas individuais de aveia. Para cada planta, um desenho da panícula principal foi desenvolvido. A partir dos desenhos obtidos nas progênies da população F2, seis classes fenotípicas distintas foram produzidas. A proporção fenotípica esperada de 3:9:4 (nuda:segregante:com casca) foi a que melhor se ajustou pelo teste do Qui-quadrado. Na geração F3, os resultados demonstraram concordância com a hipótese observada na geração F2. O caráter nuda em aveia é herdado por dois genes e a maior expressão deste caráter ocorre no terço superior das panículas. A expressão deste caráter em aveia não é completa, mesmo em genótipos homozigotos.

Abstract in English:

The aim of this study was to estimate the number of genes that control the naked (hull-less) trait and the mode of expression of this characteristic in panicles of hexaploid white oat. Parents and the segregating population (in the F2 and F3 generations) were evaluated in regard to the presence and distribution of naked grains in panicles of individual oat plants. For each plant, a drawing of the main panicle was developed. From the drawings obtained in the progenies of the F2 population, six distinct phenotypic classes were produced. The expected phenotypic proportion of 3:9:4 (naked:segregating:hulled) was that which best fit by the Chi-square test. In the F3 generation, the results showed agreement with the hypothesis observed in the F2 generation. The naked trait in oat is passed on by two genes and the greatest expression of this trait occurs in the upper third of the panicles. Expression of this trait in oats is not complete, even in homozygous genotypes.

BRS Vivi is a contribution to the expansion of the range of available cultivars on the market and the genetic basis of sweet corn in Brazil.

Abstract in English:

The development and characteristics of UFV-M7, a mutant with photoperiod insensitivity to beginning of flowering, are described. This genotype was fortuitously obtained by regeneration of nodal segments, exposed to gamma radiation. The possibility of incorporating photoperiod insensitivity into cultivars for producing regions is discussed for latitudes where the photoperiod determines the seasonality of harvest.

In the Northeast of Brazil, sugarcane cultivar RB962962 is harvested at the end of the cycle, between December and February, with a high sugar yield per area. Recommended for sandy soils of medium texture and fertility, it is resistant to the major diseases and fast-growing in plant and ratoon crops.

The cultivar CD 122 was tested in the wheat-producing regions VCU 1, 2 and 3, with an average grain yield of 3,527 kg ha-1 when fungicide-treated, exceeding the average of the control cultivars by 5%. CD 122 is resistant to leaf rust and was classified as bread wheat.
