Activity of alternative oxidase and plant uncoupling mitochondrial protein in potato tubers stored at low temperature or submitted to artificial aging Pinheiro, Hugo Alves Borges, Regis Silva, Marco Aurélio Pedron e Centeno, Danilo Cruz

Abstract in Portuguese:

Tubérculos de batata (Solanum tuberosum L. cv. Binje) foram armazenados a 18 e a 4ºC, ou envelhecidos artificialmente, com o objetivo de verificar possíveis alterações no metabolismo mitocondrial, sobretudo nas atividades da oxidase alternativa (AOX) e da proteína desacopladora de mitocôndrias de plantas (PUMP). Nenhuma alteração se observou, em qualquer tempo, nas taxas de consumo de oxigênio (OCR), controle respiratório (RC) e razão ADP/O de mitocôndrias obtidas após armazenamento a 18ºC. Já após cinco e dez dias a 4ºC, verificou-se considerável incremento em OCR, tanto no estado respiratório 3 quanto no 4, o que resultou em reduções em RC e na razão ADP/O. A atividade da AOX foi evidente após cinco dias a 4ºC, sendo maior após dez dias nessa temperatura. Houve pequenas alterações na atividade da PUMP depois de um dia de exposição a 18ºC ou a 4ºC, bem como após o envelhecimento artificial de fatias dos tubérculos. Passados cinco e dez dias de exposição a 4ºC, foi possível notar um incremento no inchamento mitocondrial, em presença de valinomicina e ácido linoléico. Apesar das alterações nas atividades da AOX e PUMP, notaram-se apenas pequenos incrementos nas atividades da peroxidase do ascorbato (APX) e catalase (CAT), maiores após cinco e dez dias de exposição a 4ºC. Os resultados revelam o efeito combinado da baixa temperatura e do tempo de armazenamento na promoção de incrementos na atividade da AOX, evidenciando um possível papel protetor dessa proteína contra estresse oxidativo mitocondrial, mediado por baixa temperatura.

Abstract in English:

Potato tubers (Solanum tuberosum L. cv. Binje) were stored at 18 and 4ºC in order to verify changes in mitochondrial metabolism, especially in the activities of the alternative oxidase (AOX) and plant uncoupling mitochondria protein (PUMP). Tubers were also sliced and submitted to artificial aging. The oxygen consumption rate (OCR), respiratory control (RC) and ADP/O ratio for mitochondria obtained after storage at 18ºC were not altered, regardless of the time of storage. Considerable increment in OCR, for both respiratory states 3 and 4, was observed after storing the tubers for five and ten days at 4ºC; and it was accomplished by reductions in RC and ADP/O. The AOX activity was evident after five days at 4ºC, and it was enhanced after ten days of storage at that temperature. No significant changes in PUMP activities were observed after one day at 18ºC or at 4ºC, neither after artificial aging. However, increased swelling of potato mitochondria was observed in the presence of valinomycin and linoleic acid when the tubers were kept at 4ºC for five and ten days. Besides the changes in AOX and PUMP activities, slight alterations in ascorbate peroxidase (APX) and catalase (CAT) activities were observed after storing the tubers at 4ºC for 5 days. The activities of these enzymes were higher after storage at 4ºC for 10 days. The results suggest the combined effect of low temperature and time of exposure in promoting AOX activity, probably as a protective mechanism against cell damage in response to mitochondrial oxidative stress.

Effects of changes in the photosynthetic photon flux density on net gas exchange of Citrus limon and Nicotiana tabacum Pimentel, Carlos Ribeiro, Rafael Vasconcelos Santos, Mauro Guida dos Oliveira, Ricardo Ferraz de Machado, Eduardo Caruso

Abstract in Portuguese:

O objetivo desse estudo foi avaliar o efeito da variação da densidade de fluxo de fótons fotossintéticos (DFFF) nas trocas gasosas de Citrus limon, uma espécie lenhosa, e Nicotiana tabacum, uma espécie herbácea. Quando a DFFF foi aumentada de 50 para 350 mimol.m-2.s-1 durante 60 min, retornando em seguida a 50 mimol.m-2.s-1, a taxa de assimilação de CO2 (A) aumentou e estabilizou após 15 min, mas a condutância estomática (g s) continuou crescendo, em ambas as espécies. Quando se retornou à baixa DFFF, A diminuiu instantaneamente nas duas espécies, mas a g s do limoeiro diminuiu lentamente durante 60 min, enquanto em tabaco, a g s levou apenas 15 min para decrescer. Essa diferença nas respostas é provavelmente devida à hipersensibilidade dos estômatos de espécies lenhosas, como Citrus, quando submetidas à alta DFFF, retardando o seu fechamento, quando de volta à baixa DFFF. Além disso, quando o limoeiro, crescendo sob DFFF de 300 mimol.m-2.s-1, foi submetido subitamente a 600 mimol.m-2.s-1, iniciou-se uma flutuação de A e g s, levando 150 min para se estabilizar. As causas dessa oscilação são discutidas em função do ajuste da eficiência intrínseca no uso de água, devido ao controle estomático.

Abstract in English:

The objective of this study was to evaluate the effects of changes in the photosynthetic photon flux density (PPFD) on net gas exchange of Citrus limon, a woody species, and Nicotiana tabacum, an herbaceous species. When PPFD was increased from 50 to 350 mumol.m-2.s-1 and returned to 50 mumol.m-2.s-1 after 60 min, the CO2 assimilation rate (A) increased and stabilized after 15 min in both species. Stomatal conductance (g s), however, continued to increase. After returning to low PPFD, A immediately diminished to a low value for both lemon and tobacco. Stomatal conductance of lemon diminished slowly over 60 min, whereas g s for tobacco took only 15 min to decrease. This difference in behavior is probably due to over-sensitivity of stomata of woody species, such as Citrus, when exposed to high light, retarding stomatal closure on return to low PPFD. Furthermore, when lemon, growing at a PPFD of 300 mumol.m-2.s-1, was submitted to a step increase of 600 mumol.m-2.s-1, there was an oscillatory behavior of A and g s requiring 150 min to stabilize. The causes of this behavior are discussed with respect to improved intrinsic water use efficiency by stomatal closure.

Germination of dormant seeds of Stylosanthes humilis as promoted by ethylene accumulation in closed environments Ribeiro, Dimas Mendes Barros, Raimundo Santos

A germinação de sementes dormentes escarificadas de estilosante (Stylosanthes humilis) é muito baixa em placas de Petri, mas pode ser considerável em frascos de Erlenmeyer selados devido a um acúmulo de etileno em sua atmosfera. Quando esse etileno foi adsorvido por solução de perclorato de mercúrio, a emanação do regulador gasoso das sementes dormentes e sua germinação foram drasticamente diminuídas. O etileno produzido pelas sementes dormentes, ao difundir-se na atmosfera dos frascos, promoveu maior acúmulo do próprio gás e um correspondente aumento da germinação. Sementes dormentes e não-dormentes não germinaram quando a biossíntese do regulador foi completamente bloqueada.

Abstract in English:

Germination of scarified dormant seeds of Townsville stylo (Stylosanthes humilis) is very low in Petri dishes, but may be appreciable in sealed Erlenmeyer flasks if ethylene accumulates in their atmospheres. When ethylene in the flask atmosphere was fixed by a mercuric perchlorate solution, emanation of the gaseous regulator and germination of dormant seeds were drastically decreased. On the other hand, ethylene produced by dormant seeds and accumulated in the flask atmosphere led to a further enhancement of ethylene accumulation and a corresponding increased seed germination. Germination of both dormant and non-dormant seeds did not occur when ethylene biosynthesis was completely blocked.

Comparative stomatal conductance and chlorophyll a fluorescence in leaves vs. fruits of the cerrado legume tree, Dalbergia miscolobium Lemos Filho, José Pires de Isaias, Rosy Mary dos Santos

O objetivo deste estudo foi avaliar, comparativamente, a condutância ao vapor d'água e a eficiência fotoquímica, utilizando medidas de fluorescência da clorofila a, em folíolos e frutos de Dalbergia miscolobium, o jacarandá-do-cerrado. A freqüência de estômatos nos folíolos foi 20 vezes superior à dos frutos, e este fato foi relacionado aos menores valores de condutância nos últimos, em comparação aos dos folíolos. O rendimento quântico potencial do PSII (Fv /Fmax) foi significativamente inferior nos frutos quando comparados aos folíolos. Os valores de Fv /Fmax para os folíolos aumentaram, durante a tarde, para acima de 0,8, indicando fotoinibição dinâmica. Para os frutos, os valores de Fv /Fmax permaneceram baixos, mesmo no início da manhã, revelando a fotoinibição crônica. As folhas apresentaram maior rendimento quântico efetivo (deltaF/F'max) e maior taxa aparente de transporte de elétrons (ETR) do que os frutos. Pode-se concluir que o pericarpo dos frutos de D. miscolobium, como outros tecidos verdes apresentam alguma atividade fotossintética, podendo, portanto, contribuir para a manutenção dos frutos e/ou para o desenvolvimento das sementes.

Abstract in English:

The aim of this study was to compare water vapor conductance and chlorophyll a fluorescence between leaflets and fruits of Dalbergia miscolobium, the Jacaranda tree. The frequency of stomata on the leaflets was 20 times higher than that observed on the fruits, and this was related with the lower conductance of the fruits in comparison with the leaflets. The potential quantum yield of PSII (Fv /Fmax) was significantly lower in fruits than in leaflets. The Fv /Fmax values for leaflets increased to over 0.8 during the afternoon, indicating the occurrence of dynamic photoinhibition. In contrast, Fv /Fmax values for fruits remained low even at early morning, indicating the occurrence of chronic photoinhibition. The maximum values of effective quantum yield (deltaF/F'm), and of the apparent electron transport rate (ETRmax) were higher in leaflets than in fruits. It was concluded that, like other green tissues, the pericarp of D. miscolobium was photosynthetically active, and therefore can contribute to the maintenance of the fruits and/or to the development of the seeds.

Isolation of enzymes involved in threonine biosynthesis from sorghum seeds Ferreira, Renato Rodrigues Vendemiatti, Ariane Meinhardt, Lyndel Wayne Lea, Peter John Azevedo, Ricardo Antunes

Abstract in Portuguese:

As sementes dos cereais são pobres em aminoácidos essenciais, principalmente lisina, triptofano e treonina. Os aminoácidos lisina e treonina são sintetizados na via metabólica do aspartato. Apesar de a maioria das enzimas da via do aspartato ter sido isolada e caracterizada em várias espécies de plantas, o metabolismo de lisina e treonina é totalmente desconhecido em sorgo. Isolamos duas enzimas, aspartato quinase (AK) e homosserina desidrogenase (HSDH) de sorgo, estabelecendo condições ótimas de ensaio para determinação da atividade de AK e HSDH. Observaram-se as atividades mais elevadas de AK e HSDH em sementes imaturas. A atividade de AK foi inibida por treonina e lisina, indicando a existência de duas isoenzimas, uma sensível à inibição por treonina e, a outra, sensível à inibição pela lisina, predominando, esta última, nas sementes de sorgo. A atividade de HSDH foi inibida por treonina, indicando a existência de uma isoenzima sensível à treonina, a maioria da atividade, entretanto, não foi inibida por treonina, sugerindo a existência de uma segunda isoenzima de HSDH predominante resistente à inibição pela treonina.

Abstract in English:

Cereal seeds are poor in essential amino acids, particularly lysine, tryptophan and threonine. The amino acids lysine and threonine are synthesized in the aspartate pathway. Although most of the enzymes of the aspartate pathway have been isolated and characterized in higher plant species, the metabolism of lysine and threonine is totally unknown in sorghum. We have isolated two enzymes, aspartate kinase (AK) and homoserine dehydrogenase (HSDH) from sorghum. Optimum assay conditions were established for the determination of AK and HSDH activities. The highest level of activity was observed in immature seeds. AK was shown to be inhibited by threonine and lysine indicating the existence of at least two isoenzymes, one sensitive to threonine inhibition and the other sensitive to lysine inhibition with the latter being predominant in sorghum seeds. HSDH was shown to be inhibited by threonine indicating the existence of a threonine-sensitive HSDH, however, most of the activity was not inhibited by threonine, suggesting the existence of a second predominant isoenzyme of HSDH resistant to threonine inhibition.

EgLFY, the Eucalyptus grandis homolog of the Arabidopsis gene LEAFY is expressed in reproductive and vegetative tissues Dornelas, Marcelo Carnier Amaral, Weber A. Neves do Rodriguez, Adriana Pinheiro Martinelli

O gene EgLFY, clonado de Eucalyptus grandis, possui homologia de seqüência com o gene de identidade meristemática LEAFY (LFY) de Arabidopsis e FLORICAULA (FLO) de Antirrhinum. EgLFY é, preferencialmente, expresso nos órgãos florais em desenvolvimento de eucalipto, obedecendo a um padrão similar ao descrito para o gene LFY de Arabidopsis. Experimentos de hibridização in situ mostraram que o gene EgLFY é fortemente expresso em meristemas florais no início de seu desenvolvimento e, então, sucessivamente, durante a formação dos primórdios de sépalas, pétalas, estames e carpelos. Há expressão também nos primórdios foliares de árvores adultas. A expressão da região codificadora de EgLFY sob o controle do promotor do LFY de Arabidopsis pôde complementar mutantes lfy nulos em plantas de Arabidopsis transgênicas. Essas observações sugerem que o gene EgLFY possui um papel similar ao de LFY no desenvolvimento floral e que os mecanismos básicos envolvidos na iniciação e no desenvolvimento floral em Eucalyptus podem ser semelhantes aos de Arabidopsis.

Abstract in English:

The EgLFY gene cloned from Eucalyptus grandis has sequence homology to the floral meristem identity gene LEAFY (LFY) from Arabidopsis and FLORICAULA (FLO) from Antirrhinum. EgLFY is preferentially expressed in the developing eucalypt floral organs in a pattern similar to that described previously for the Arabidopsis LFY. In situ hybridization experiments have shown that EgLFY is strongly expressed in the early floral meristem and then successively in the primordia of sepals, petals, stamens and carpels. It is also expressed in the leaf primordia of adult trees. The expression of the EgLFY coding region under control of the Arabidopsis LFY promoter could complement strong lfy mutations in transgenic Arabidopsis plants. These data suggest that EgLFY plays a similar role to LFY in flower development and that the basic mechanisms involved in flower initiation and development in Eucalyptus may be similar to those occurring in Arabidopsis.

Changes in growth and superoxide dismutase activity in Hydrilla verticillata L. under abiotic stress Panda, Sanjib Kumar Khan, Mohammad Hussain

Estudaram-se os efeitos de alguns metais pesados (cromo, zinco, cobre e cádmio), da temperatura, da salinidade induzida por NaCl e do estresse hídrico induzido por PEG sobre o crescimento e a atividade da dismutase do superóxido (SOD) em Hydrilla verticillata L. Quase todos esses metais reduziram o crescimento, mas tiveram efeito modulador em baixas concentrações. Os tratamentos de salinidade induzida por NaCl, estresse hídrico e temperatura, também o reduziram. A atividade do SOD foi inibida por cromo e zinco, mas aumentada por cobre e cádmio. O estresse salino aumentou a atividade do SOD, enquanto o hídrico o diminuiu. O SOD mostrou-se termoestável, sendo a maior atividade atingida a 45ºC.

Abstract in English:

A study was made of the role of various heavy metals (chromium, zinc, copper and cadmium) together with the effects of NaCl-salinity, PEG-mediated water stress and temperature on growth and superoxide dismutase (SOD) activity in hydrilla (Hydrilla verticillata L.). Almost all metals studied reduced growth but presented a modulatory effect at lower concentrations. NaCl-salinity, water stress and temperature treatments also reduced growth. SOD activity was inhibited by both chromium and zinc but increased by copper and cadmium. NaCl-salinity increased SOD activity while water stress decreased it. SOD showed thermostability with a higher activity being recorded at 45ºC.
